Growth Regulation of A431 Cells MODULATION OF EXPRESSION OF TRANSFORMING GROWTH FACTOR-a mRNA
نویسنده
چکیده
To explore bidirectional regulatory interactions between interferons and autocrine polypeptide factors, we examined the modulation of expression of transforming growth factor-a and 2’,5’-oligoadenylate synthetase activity in A431 epidermoid carcinoma cells after treatment with interferon-y and transforming growth factor-a. Treatment of A431 cells with interferon-y increased steady state levels of transforming growth factor-a mRNA by 4-fold and increased the levels of transforming growth factor-a in the culture medium. There were additive growth inhibitory effects upon coaddition of exogenous transforming growth factor-a and interferon-y to the cultures. Addition of transforming growth factor-a to A431 cell cultures in the absence of interferon could stimulate the induction of 2’,5’-oligoadenylate synthetase activity by more than a-fold. These findings demonstrate that the induction of transforming growth factor-a in interferony-treated A431 cells could act to regulate interferoninduced gene(s), e.g. 2’,5’-oligoadenylate synthetase, suggesting interactions between a potential autocrine growth factor and the interferon system in the growth regulation of A43 1 cells.

Transforming growth factor betas (TGF?s) are one of the most widespread and versatile cytokines. The three mammalian TGF? isoforms, ?1, ?2, and ?3, and their receptors regulate proliferation of neuronal precursors as well as survival and differentiation in neurons of developing and adult nervous system. Functions of TGF?s has a wide spectrum ranging from regulating cell proliferation and differ...

Background: During antigen capture and processing, mature dendritic cells (DC) express large amounts of peptide-MHC complexes and accessory molecules on their surface. DC are antigen-presenting cells that have an important role in tolerance and autoimmunity. The transforming growth factor-beta1 (TGF-β1) cytokine has a regulatory role on the immune and non-immune cells. The aim of this study is ...

Transforming growth factor betas are multifunctional polypeptides in the cytokine superfamily. They have a growth inhibitory role on hemopoietic progenitor cells in semisolid colony assay as well as in long-term bone-marrow culture. TGF - β2 represses stromal cells, stem cell factor gene transcription, and decreases the stability of c-kit transcripts in hemopoietic cells. TGF-β also modulat...

Transforming growth factor beta (TGFβ1, β2, and β3) are 25 kDa disulfide-linked homodimers that regulate many aspects of cellular functions, consist of proliferation, differentiation, adhesion and extracellular matrix formation. TGFβs mediate their biological activities by binding of growth factor ligand to two related, functionally distinct, single-pass transmembrane receptor kinases, known as...
